Direct estimate of microbiome size

Through the combination of results from next generation sequencing with qPCR based direct estimates of random fragment copy numbers we were able to estimate the total size of metagenome of an experimental soil microbiota. The size was about 11 log nucleotides, corresponding to about 20 000 bacterial genomes. Our approach will be useful preparing large scale metagenome sequencing projects.
